The Eswatini rivals, Nsingizini Hotspurs, capitalized on the blunder to win the shootout 4-1 at the Prince Mangosuthu Buthelezi Stadium.
After the match ended in a tense goalless draw, the pre-season tournament fixture skipped extra time and went straight to penalties. Mpisane stepped up as one of the designated penalty takers for the Highlanders.
However, the high-profile son of Shauwn Mkhize completely missed the target after an elaborate stutter-step approach, leaving him empty-handed as Nsingizini secured their spot in the final.
Reigning Eswatini treble-winners Nsingizini Hotspurs now advance to face amateur side Ulundi United in the grand final, scheduled for a 16:00 kick-off.
Ulundi United booked their place in the showcase match earlier on Saturday morning by stunning Betway Premiership giants AmaZulu FC with a comprehensive 2-0 victory.
